Poster: Large Insert Libraries
Since the URGV (Unite de recherche en genomique Vegetale) have been set-up at INRA two years ago, we have constructed several BAC libraries from different cultivated plants, important to France and Europe. These include: a hexaploid wheat (Triticum aestivum) BAC library of 1000 000 clones that is 7X haploid genome equivalent with an average insert size of 140 Kb; an oilseed rape (Brassica napus) BAC library of 75000 BAC clones that is 12 X haploid genome equivalent and 200 Kb average insert size; Three Grapevine (Vitis vinifera) BAC libraries (from three different cultivars) of about 70 000 BAC clones each and 150 Kb of mean insert size. Other BAC libraries from other cultivated plants or cultivars are being constructed. Methods for obtaining these BAC libraries (conditions, vectors…) as well as their utilization in genome organization studies and genes and QTLs cloning will be presented.
