ORGANISM STRAIN:
Magnaporthe grisea, strain 70-15
BRIEF SUMMARY OF SEQUENCING STRATEGIES:
Genomic sequencing of chromosome 7 (4.2Mb) - ~5 X sequence
coverage of 41 BAC clones representing the minimum tiling path
of chromosome 7 will be accomplished. BAC clones will be
sheared to ~2 kb fragments and cloned in a plasmid vector.
Sequence will be obtained from both ends of ~815 clones per BAC. After vector and contaminating sequences are removed, sequence will be assembled into contigs.
EST sequencing - A total of 35,000 ESTs will be produced from a
minimum of 8 cDNA libraries prepared from different stages of
growth and development as well as from cells subjected to
various stress conditions. Three to four thousand ESTs will be
generated from the 5'end per library for a total of 30,000
reactions. From these, a set of ~5,000 ESTs representing
unique genes will be sequenced from the 3' end.

All data will be maintained in a central database available
through www.cals.ncsu.edu:8050/fungal_genomics/ The data will
be incorporated into a database housing other physical and
genetic data as well as BAC-end sequence data developed
previously.
Frequency of Data Release & Updates:
BAC sequencing ÿ consensus sequence for each completed BAC will
be deposited in GenBank by FTP immediately after alignments
have been checked manually. Each BAC clone sequence will be
updated a minimum of twice. First following annotation based
on ORF prediction and GenBank Blast matches. A second update
will include matches to rice blast ESTs and will occur after
EST sequencing is completed.
EST sequencing ÿ ESTs (with vector sequence removed) will be
deposited in GenBank by FTP on a weekly basis. If homologs
with known function are identified by Blast alignments, the a
ppropriate annotation will be added to the record.
