ORGANISM STRAIN:
Mannheimia (Pasteurella) haemolytica serotype A1 Strain PHL213 = SH789

SUMMARY OF SEQUENCING STRATEGY:
DNA comprising the entire genome of strain PH213 will be used to create small M13 (2-4 kb) and large cosmid (20-30 kb) libraries. M13 libraries will be used for random shotgun sequencing (ca. 30,000 reads) using dye terminator chemistry and capillary electrophoresis. Cosmid library members (ca. 1000 each) will be end-sequenced and sequences used to create an ordered cosmid maps, or scaffolds, on which to array the randomly generated sequences. Sequence assembly will be performed by using PHRED and PHRAP. Gene clusters, structural features, and open reading frames will be identified and annotated using a number of computational methods, including BLAST-BEAUTY analysis. Annotated sequences will be organized and released on a new microbial genomes website at the Baylor College of Medicine Human Genome Sequencing Center.

Strains will be maintained in the laboratory of Dr. Highlander at Baylor College of Medicine. Our preference will be to distribute large insert plasmids, but small insert plasmids created for the shotgun phase of sequencing may also be made available.
