In order to understand the genetic control mechanisms involved in the symbiotic nitrogen fixation, we have initiated genome sequening of Mesorhizobium loti MAFF303099, a symbiont of Lotus japonicus.
The genome of M. loti MAFF303099 consists of three circular replicons: a chromosome of 7.2 Mb and two plasmids of 360 Kb and 200 Kb long. An average GC content of the genome was estimated to be 61.7%. The nucleotide sequence of the genome was determined by the whole genome shotgun method. The random sequences were accumulated until seven folds of the genome size were covered, which were assembled into 370 sequence contigs after assembly. These contigs were then assigned to the genome in order, and the genome size was estimated to be 7,589 Kb. As of Oct. 13 2000, most of the sequence gaps were filled, and the sequence editing is under way (30% completed). The latest status of the project will be presented.
