Large insert libraries such as BAC and YAC play an important role in the global physical mapping of genomes, their structural and functional comparisons and the identification of genes. Very few such libraries have so far been reported for plant pathogenic fungi. Fusarium solani f.sp. glycines is the causative agent of sudden death syndrome (SDS) of soybean and results in severe crop losses in the Midwestern United States. Here we report the construction of a BAC library containing 12,288 clones with an average size of 110 kb. The library was constructed in a binary vector (pCLD04541) that we have earlier reported in the construction of soybean BAC libraries. The vector is readily usable in plant transformation, therefore, the library will be very useful in determining the genomic regions of the pathogen involved in resistance and susceptible response of the soybean. We also developed a new fluorescent labeling based BAC fingerprinting technique using CEQ fragment analysis system. A large contig constructed from BAC fingerprints will also be reported in the poster.
