AA GENOME SPECIFIC PROMOTER LIKE DNA SEQUENCE FROM INDICA RICE (IR36) GENOME.

Rice genome is small enough (0.6 pg) for molecular genetic studies particularly in the background that enough data have been accumulated with the initiative of rice genome projects. We have cloned a 700 base pair Mbo I repeat from IR36 rice genome which is non-ribosomal in nature. During characterisation and sequencing the clone (pR36VII) shows the presence of TATA sequence followed by a ATG codon. A significant portion of the clone thus represents a putative promoter sequence. Restriction analysis shows recognition sequences for 13 restriction endonucleases. The distribution pattern of this clone indicates that the said sequence is AA genome specific and is absent from Aeries officinalis, (CC) genome. Also our data suggest that the same could be further dissected for a successful RFLP probe. The copy number of this sequence indicates it as a middle repetitive DNA sequence.
