Brachypodium distachyon, with a genome size of ~300 Mb, is an emerging model system for temperature grasses and bioenergy crops. To facilitate genomic studies in Brachypodium we used the Illumina (Solexa) microread sequencing platform to deeply sample a collection of cDNA libraries representing a diverse array of tissues, treatments and developmental stages. These data enable the efficient identification of transcription units and provide a comprehensive view of the Brachypodium transcriptome structure, composition, and organization, which will increase the accuracy and coverage of genome annotation efforts.
