GeneMaker is a novel, automated gene synthesis platform that can produce any gene with perfect accuracy regardless of sequence complexity or length. Blue Heron Bio’s proprietary design software automatically creates an optimal build strategy and stores the resulting assembly instructions and parts list in an Oracle database. Double-stranded oligos (“duplexes”) are made to the specifications designated by the design software and loaded onto Blue Heron Bio’s proprietary gene assembly instrument. This instrument sequentially adds “duplexes” to a solid matrix within a proprietary column to assemble a specific DNA fragment. The advantages of this assembly method are three fold: 1) it can be used to assemble any length DNA fragment, from 50 base pairs to 40,000 base pairs or longer; 2) the sequential addition avoids errors of order that commonly occur during PCR synthesis, and 3) the automation enables high throughput, highly accurate gene production at low cost with quick turnaround time.
