Parallab™ Technology: Miniaturized Cycle Sequencing Reactions

The Life Sciences Group of Brooks Automation, Inc. has developed a fully automated, integrated platform to perform nanoliter volume cycle sequencing reactions. Our standard DNA cycle sequencing reaction is performed in a total volume of 500 nanoliters, which is 98% smaller than a standard reaction. A key element in the Parallab 350™ is the proprietary Nano-Pipetter. This innovative device utilizes 96 glass tubes to perform all of the reaction processing steps, including: assembling, mixing, thermal cycling, and magnetic bead purification. After each sample set is completed, the Nano-Pipetter is cleaned and reused, greatly reducing the amount and cost of consumables normally associated with conventional processing methods. In this presentation, we discuss sequencing data from a wide range of conditions. The Parallab 350 produces quality sequencing data from plasmids, PCR products, BAC’s and TempliPhi™. Because the reactions are miniaturized, not diluted, we have proven success with a 16-fold concentration range of input template. We also show a reduction in Big Dye® reagent used to 160th of a standard 1x reaction. In addition, integrated tools for magnetic bead dye terminator removal allow the automation of multiple commercially available kits. This unique combination of attributes (nanoliter volume reactions, reduced consumables, and complete automation) enable researchers to easily integrate this highly novel platform into a variety of laboratory environments, for a broad number of applications and immediately see cost and labor savings without compromising data quality.
