CGAT (after Citrus Genome Analysis Team) of NIFTS/NARO JAPAN has been focusing to investigate genes and genome structure basically related to fruit quality of Satsuma mandarin (C. unshiu Marc.). For the purpose, the EST cataloguing of cDNA libraries from fruit tissues has been performed and so far near 10,000 EST clones have been analyzed. The sequence of EST has been applied to generate co-dominant CAPS markers and QTLs related to fruit quality such as acidity, seedlessness, fruit color have been mapped on the CAPS linkage groups. These markers will be used for MAS for breeding and description of cultivar genotyping. To proceed the functional genomics on fruit physiology, cDNA microarrray is preparing to profile the expression pattern of ESTs during fruit development and maturation. So far, by using chip with 3,000 independent clones, some clones were selected to isolate the candidate of specific promoters. The model system will be prepared to induce precocious flowering and fruiting by genetic transformation for the identification of gene function of the selected clones.
