Poster: Genome Sequencing & ESTs
Two bovine BAC clones containing BoLA class IIB sequence were found to contain the locus BoLA DIB1.
Sequencing of these two BACs revealed alternate alleles for the DIB1 locus. Sequence comparison to a partial DIB1 (GenBank Acc. U69097) sequence published by Stone et al showed 100% similarity in the coding regions. DIB1 gene structure for each BAC was predicted using GeneScan ( http://genes.mit.edu/GENSCAN.html ). Both predicted CDS and protein sequences matched the published sequence 100%. The remaining predicted exons were divergent between the two clones. One allele of DIB1 had DNA inserted into each of three introns compared to the alternative allele. These three inserts contained 94 bases, 955 bases and 760 bases, respectively. The predicted divergence of the exons appeared to occur in the cytoplasmic tail.
