THE SWEDISH POPULUS EST PROJECT

The Swedish functional genomics tree programme consisting of groups at the Royal institute of Technology, Stockholm and Umeå Plant Science Centre will at this meeting present more than 60 000 EST sequences obtained by sequencing more than 14 tissue specific cDNA libraries from Populus tremula x tremulodies. Based on this EST collection, a unigene set consisting of ca 13 000 sequences has been used for production of DNA micro arrays. Our global gene profiling approach is linked to protein and metabolite profiling through large-scale proteomics and metabolomics. Our vision is that this genomic approach will allow an unprecedented and comprehensive insight into gene expression correlated with the amounts and types of proteins and metabolites present in specific tissues. Eventually this will result in the development of a "blueprint" of the tree cell that can serve as a foundation for elucidating the mechanisms of wood formation and tree development We will also show the utility of combining poplar transcript profiling with a rapid analysis of gene function in Arabidopsis. Finally, we will also show how target selection through tissue-specific transcript profiling in Populus can be combined with a large scale characterization of gene function in transgenic trees.
