A SIMPLE 96 WELL METHOD FOR THE PURIFICATION OF BAC TEMPLATES.

We have developed a simple method for purifying BAC templates in a 96 well format using a modified Ion exchange resin for nucleic acid binding. The plate can be used with a vacuum manifold or centrifuge for binding and elution. Yields average between 0.8 and 1.0 ug from a 5 mL overnight culture (Terrific Broth). High quality sequence reads of up to 700 bases have been obtained. A discussion of the protocol and the quality of results obtained using the method is presented.
