Phytophthora root and stem rot is an important disease of soybean. It is caused by the oomycete pathogen Phytophthora sojae. A series of Rps genes confer resistance of soybean against P. sojae. We have applied a map-based cloning approach in isolating the most widely used gene, Rps1-k. We applied AFLP mapping and BAC cloning technologies in developing a BAC contig for the Rps1-k region. Screening of several BAC libraries representing over 20 genome equivalents showed that clones representing this region are highly underrepresented in the BAC libraries. Genetic and physical mapping data indicated that recombination is suppressed in this region. We have observed ~1.5 mb DNA/cM in this region. The Rps1-k region was localized to ~175 kb DNA. There are at least nine copies of coiled coil-NBS-LRR-type sequences in this region. Currently, the entire Rps1-k region is being sequenced. Complementation experiments are also being carried out.
